With my UMIK-1 plugged in, this MiniDSP UMIK-1 detected box doesn't pop up when I open REW.

Any suggestions on how to upload my calibration file?

Are you running the latest Beta file from REW(at least 5.01 beta 14)? Most likely the reason I guess. Just double check in case.
You can load the calibration from the preferences section of REW.

I am running the latest version of the REW software.

This worked. I went to "preferences", then clicked "preferences" again in the drop down menu. Then clicked on the "Mic/Meter" tab. Here, you can click "browse", find your calibration file, and upload it.

Merely curious, as I've been having similar issues, which version of windows are you running?
