Bought a brand new Asus laptop with windows 10, downloaded Java update and downloaded the minidsp 2x4HD plug in.
Keep getting the “cannot connect to minidsp, please plug in board” window.
Everything is plugged in how it should be so what the heck is going on?

Try to turn off windows defender during the installation, this might be the reason why it puts it in quarantine and block the access. Usually when you run windows 10, there will be automatically updated, but maybe you didn't get them for some reason. I am a medical student, and when I got a new laptop after consulting this website thecursedcrusade.com/best-laptop-for-nursing-student/ , I installed all the updates and drivers on windows 10, because there were missing a few, and so I will avoid similar errors in the future.
