I2S stands for Inter IC sound and is a way to send digital audio between IC's. While it's not very complex to understand, it's there for those of you who know how it works and give DIYers the opportunity to interface with their existing DIY stuff (e.g. a good preamp) You may want to have a look at our I2S technical note on the download section of our site for some details.
Now, I2S is not intended for long distances (i.e. over a cable from a PC sound card). You could if you know a bit of electronic maybe tweak your PC to grab the I2S from a motherboard, but it means too much work/knowledge involved and would not recommend it if you don't know what you are doing. USB audio is indeed the way to go when you have a digital audio PC source. There are however some issues with getting a format that the DSP can work with (sample rate converter needed).
Getting a nice 24bit USB audio solution for multi-channel is a design we already made as a quick sketch (no proto/no full engineering). If we do it, we'll do it right so it won't be an overnight 16bit DAC solution as it's readily available. There are also a couple of other cards that may go before this one (e.g. high fidelity audio) though, and we'll have to first establish which one are worth doing it.
